چکیده

The renal glomerulus is composed of three types of glomerular cells (mesangial cell (MC), endothelial cell and podocyte) and extracellular matrix (ECM) consisting of the glomerular basement membrane (GBM) and mesangial matrix. It constitutes ahighly specialized microcirculation in which the permeability characteristics of the capillary wall allow its unique filtrationfunction.TheproliferationofMCs,anincreaseofmesangialECMand detachment podocyte from GBM are key biological features ofprogressiveglomerulonephritis (GN), leading to glomerularscarring and dysfunction. Thus, thestudy of themolecularandcellularmechanisms responsible for pathological glomerular alterations may help to elucidate the pathogenesis of progressive glomerular diseases. A growing body ofevidenceindicates thatβ1 integrin family (β1 integrins), that mainly mediates cell adhesion to ECM, controls cell behaviors such as cell migration, proliferation, apoptosis and ECM assembly. In addition, a correlation between glomerular expression of β1 integrins and their ligand ECM components is observed in various human and experimental GN, suggesting that altered β1 integrins-mediatedcellbehaviorsmay contribute to the progression of GN. It is now becoming apparent that the expression of glomerular β1 integrins is not only critical for maintaining the glomerularcapillary permeability but it modulates cell signalingpathways regulatingthecellphenotypes involvedintheprogression of glomerular diseases. Ovarian Stimulation by Exogenous Gonadotropin Decreases the Implantation Rate and Expression of Mouse Blastocysts Integrins

Background: Integrins are heterodimeric glycoprotein receptors that regulate the interaction of cells with extracellular matrix and may have a critical role in implantation. The aim of this study was to investigate the effect of ovulation induction on the expression of α4, αv, β1, and β3 integrins in mouse blastocyst at the time of implantation. Methods: The ovarian stimulated and non-stimulate...

Podocyte-associated talin1 is critical for glomerular filtration barrier maintenance.

Podocytes are specialized actin-rich epithelial cells that line the kidney glomerular filtration barrier. The interface between the podocyte and the glomerular basement membrane requires integrins, and defects in either α3 or β1 integrin, or the α3β1 ligand laminin result in nephrotic syndrome in murine models. The large cytoskeletal protein talin1 is not only pivotal for integrin activation, b...

Upregulation of α3β1-Integrin in Podocytes in Early-Stage Diabetic Nephropathy

Background. Podocyte injury plays an important role in the onset and progression of diabetic nephropathy (DN). Downregulation of α3β1-integrin expression in podocytes is thought to be associated with podocyte detachment from the glomerular basement membrane, although the mechanisms remain obscure. To determine the mechanism of podocyte detachment, we analyzed the expression levels of α3β1-integ...

Podocyte-specific RAP1GAP expression contributes to focal segmental glomerulosclerosis-associated glomerular injury.

Injury to the specialized epithelial cells of the glomerulus (podocytes) underlies the pathogenesis of all forms of proteinuric kidney disease; however, the specific genetic changes that mediate podocyte dysfunction after injury are not fully understood. Here, we performed a large-scale insertional mutagenic screen of injury-resistant podocytes isolated from mice and found that increased expres...
